چکیده

Introduction Myelin water imaging has been successfully used to assess myelin content in human and animal CNS tissue [1,2]. It has been shown that fractional amounts of water trapped between myelin sheaths – the so called myelin water fraction (MWF) [3] correlates well with the amount of myelin in a rat model of spinal cord injury (SCI) [2] and can therefore be an important marker of the cord pathology. One of the limitations of the current CPMG based myelin water imaging technique is that it is a single slice technique. It has been shown that a 3D version of this technique can be achieved by applying a second phase encoding gradient to provide localization along the slice direction [4]. In this pilot study we applied the 3D CPMG sequence to acquire very high spatial resolution 3D MWF maps from excised rat spinal cord. The 3D technique produced high quality data that compared favourably with the 2D measurements.

In vivo myelin water imaging in rat spinal cord

Introduction Measuring myelin content in a rat model of spinal cord injury (SCI) can be an important marker of the cord pathology and can be used to monitor potential SCI therapies. Myelin water imaging has been shown to measure myelin content in normal and diseased brain and spinal cord tissue [1, 2]. This technique was successfully applied in excised rat spinal cords [3], however the requirem...

High b-value q-space diffusion MRI in myelin-deficient rat spinal cords.

In this study, we explore the effect of the lack of myelin on the diffusion characteristics and diffusion anisotropy obtained from high b-value q-space diffusion-weighted MRI (q-space DWI) in excised rat spinal cords. Twenty-one-day-old myelin-deficient (md) mutant (N=6) and control rats (N=6) were used in this study. The MRI protocol included multi-slice T(1), T(2), proton density (PD) MR imag...

Ex vivo myelin water and DTI measurements of SKP-SC transplanted cell therapy in contused rat spinal cord: correlation with histology

Introduction Schwann cells differentiated from skin-derived precursors (SKP-SCs) have been shown to promote histological and functional recovery in a contusion model of rat spinal cord injury [1]. Cellular bands of SKP-SCs bridge the lesion site which promote axon growth ensheathed by peripheral (Schwann cell) myelin of endogenous as well as transplant origins. In this work, we assess the myeli...

Experimental spinal cord injury: imaging the acute lesion.

In order to obtain high resolution images of fixed excised rat spinal cords we have developed a technique using a 6-mm bore, two-turn saddle coil, with a usable imaging length of approximately 4 cm. MR imaging is performed on a prototype 31-cm bore, 1.9-T system with a 1.5-mm section thickness and 7.6-mm field of view.
